産品簡介:

    彩虹預染超低(dī)分子量蛋白(bái)質Marker可以直接觀察蛋白(bái)電(diàn)泳及清晰判斷Western Blotting的轉膜效果。本産品包含3種多(duō)肽和3種低(dī)分子量蛋白(bái)質組成,分子量範圍為(wèi)1.2kD-45kD,分子量大小(xiǎo)為(wèi)  1.2kD4.6kD10kD16kD27kD45kD (注:蛋白(bái)大小(xiǎo)已在18%Tricine-SDS-PAGE中用非預染蛋白(bái)Marker标定過。




● 使用說明:

第一(yī)次收到(dào)該産品,常溫融化後,徹底混勻,離心快甩将溶液完全收集到(dào)管底

一(yī). 制膠:

I 配制分離膠

1.按照(zhào)表一(yī)将不同體積的雙蒸水(shuǐ)、40% PAA19:1)、4×凝膠緩沖液(貨号:GB010)和乙二醇加入到(dào)小(xiǎo)燒杯中混合。

2.加入10%APSTEMED,立即混勻5-10秒(miǎo),以使溶液充分混勻。

3.在凝膠模具中迅速灌入适量分離膠溶液,然後在分離膠溶液上(shàng)輕輕覆蓋一(yī)層1 cm的無水(shuǐ)乙醇,使凝膠表面保持平整。

4.靜(jìng)置15-30分鍾,待分離膠和乙醇層之間出現一(yī)個(gè)清晰的界面表示凝膠已聚合。

II 配制濃縮膠

去除覆蓋在分離膠上(shàng)的乙醇層,用濾紙(zhǐ)将殘留乙醇吸去。

1.按照(zhào)表一(yī)将不同體積的雙蒸水(shuǐ)、40%PAA(29:1)和4×凝膠緩沖液加入到(dào)小(xiǎo)燒杯中混合。

2.加入10%過硫酸铵和TEMED,立即混勻5-10秒(miǎo),以使溶液充分混勻。

3.将梳子插入凝膠内,避免産生(shēng)氣泡。

4.靜(jìng)置30-60分鍾裝待凝膠聚合

. 電(diàn)泳:

1. Marker樣品,充分混勻後備用。不要加熱處理。

2. 電(diàn)泳前,将10×陽極緩沖液(Cat No:AB080)和10×陰極緩沖液(Cat No:CB010)用蒸餾水(shuǐ)稀釋成1×緩沖液備用。将電(diàn)泳槽的外槽加入1×陽極緩沖液,内槽加入1×陰極緩沖液,輕柔拔出梳子,将Marker或蛋白(bái)樣品(已經過2×Tricine上(shàng)樣緩沖液[Cat No:TP050]處理)加入點樣孔,參考下(xià)表進行電(diàn)泳,至每條帶都清晰可見(jiàn)時停止電(diàn)泳。整個(gè)電(diàn)泳過程大約需要2-3個(gè)小(xiǎo)時。

. 染色

根據常規實驗步驟,用配方6和7(表三)進行染色和觀察。如果使用配方6進行染色時效果不好或考慮其毒性,請選擇本公司的FastBlue蛋白(bái)染色液(貨号:RTD6202),該産品除具有染色快,無毒,靈敏性高(gāo)等特點外,還(hái)能(néng)對小(xiǎo)肽在染色中起到(dào)固定作用,不至于小(xiǎo)肽在染色和脫色中從(cóng)凝膠中脫離,是常規染色液的理想替代品。
