植物(wù)RNA提取試劑
産品簡介:
RNAplant試劑可從(cóng)植物(wù)組織,特别是富含多(duō)酚或澱粉的植物(wù)組織中(如馬鈴薯塊莖,白(bái)松松針或嫩苗,和西(xī)紅(hóng)柿葉等)提取高(gāo)純度的總RNA。
每100 ml可處理100 mg組織200次。
保存條件(jiàn):室溫運輸,4℃保存,可保存6-12個(gè)月(yuè)。
準備試劑:液氮,研缽,無RNase離心管,5 M NaCl(NC1050F 5M 氯化鈉 RNase-free,DNase-free),氯仿,異丙醇,75%乙醇(無RNase水(shuǐ)配制),無RNase水(shuǐ)。
小(xiǎo)規模提取操作步驟:(樣品<0.1 g ,使用小(xiǎo)規模提取方法):
1. 取不多(duō)于0.1 g植物(wù)組織,在液氮中研碎,轉移到(dào)1.5 ml 離心管中,加0.5 ml提取試劑,振蕩至徹底混勻。
注:振蕩不要太劇烈,否則容易導緻基因組DNA污染。
2. 室溫放(fàng)置5分鍾。
3. 4℃ 12, 000 rpm離心2-3分鍾,上(shàng)清轉入新的無RNase離心管。
4. 上(shàng)清中加入0.1 ml 5 M NaCl(NC1050F 5M 氯化鈉 RNase-free,DNase-free),溫和混勻。
5. 加入0.3 ml 氯仿,上(shàng)下(xià)颠倒混勻。
6. 4℃ 12,000 rpm離心10-15分鍾,取上(shàng)層水(shuǐ)相(xiàng)轉入新的無RNase離心管。
7. 加與所得水(shuǐ)相(xiàng)等體積的異丙醇,混勻,室溫放(fàng)置10分鍾。
8. 4℃ 12,000 rpm離心10-15分鍾。棄掉上(shàng)清,注意不要倒出沉澱。加1 ml 75%乙醇沖洗沉澱。(沉澱可能(néng)很難看(kàn)見(jiàn),應小(xiǎo)心操作。)
9. 4℃ 5000-7000rpm離心3分鍾。倒出液體,注意不要倒出沉澱。剩餘的少量液體短暫離心,用槍頭吸出,室溫晾幹1-2分鍾至沉澱幹燥。
10. 加适量無RNase水(shuǐ)充分溶解RNA,-70℃保存。
注意事(shì)項:
RNAplant有刺激性味道,操作時請在通(tōng)風處進行。
